Overview

NY-ESO-1b peptide is a synthetic cancer-testis antigen-derived peptide vaccine designed to stimulate an immune response against tumors expressing the NY-ESO-1 antigen. The NY-ESO-1 protein (also known as CTAG1B) is normally expressed only in germline tissues such as testis and placenta but is aberrantly expressed in various cancers, including ovarian, lung, breast, melanoma, and esophageal cancers. The vaccine aims to induce both humoral and cellular immunity—specifically activating CD8+ cytotoxic T lymphocytes and CD4+ helper T cells that recognize tumor cells presenting the NY-ESO-1 epitope via HLA-A2 or other MHC molecules. Clinical studies have shown that vaccination with the NY‑ESO‑1b peptide can elicit robust immune responses with minimal toxicity in patients with high-risk or advanced cancers expressing this antigen[6][10]. It is typically administered subcutaneously along with adjuvants such as Montanide ISA‑51 and CpG 7909 to enhance immunogenicity[6]. The primary goal of therapy is to prolong remission or reduce tumor burden by harnessing the patient’s own immune system.
