NY-ESO-1b peptide + CpG 7909 + Montanide ISA-51 is a combination cancer vaccine formulation designed to stimulate an immune response against tumors expressing the NY-ESO-1 antigen. The NY-ESO-1b peptide is derived from the New York esophageal squamous cell carcinoma 1 (NY‑ESO‑1) protein, a highly immunogenic cancer-testis antigen expressed in various malignancies but not in most normal tissues[2][5][7]. The vaccine includes CpG 7909, a synthetic oligodeoxynucleotide that acts as an immunostimulatory adjuvant by activating Toll-like receptor 9 (TLR9), thereby enhancing both innate and adaptive immune responses. Montanide ISA‑51 serves as an oil-based adjuvant to further boost the immunogenicity of the vaccine by promoting sustained antigen release and local immune activation[6][7]. This combination aims to induce robust CD4+ and CD8+ T cell responses targeting tumor cells presenting NY‑ESO‑1 peptides via MHC class I and II pathways. It has been investigated primarily for use in solid tumors such as ovarian cancer, melanoma, synovial sarcoma, and other cancers with documented NY‑ESO‑1 expression[6][7].
