NY-ESO-1b peptide vaccine is a therapeutic cancer vaccine developed by the Ludwig Institute for Cancer Research. It is composed of peptides derived from the NY-ESO-1 (New York esophageal squamous cell carcinoma 1) protein, also known as cancer/testis antigen 1B (CTAG1B). This antigen is a prominent target in immunotherapy because it is highly expressed in various malignancies—including melanoma, ovarian cancer, and lung cancer—while remaining largely absent in normal adult tissues except for germ cells. The vaccine is designed to stimulate the host's immune system to recognize and destroy tumor cells by priming and expanding antigen-specific CD8+ and CD4+ T cells. In clinical development, it has often been administered in combination with immunological adjuvants such as Montanide ISA-51 or TLR7/8 agonists like resiquimod to enhance the vaccine's potency and overcome tumor-induced immunosuppression.
