Overview

NY-ESO-1b.A2 is a synthetic HLA-A2-restricted peptide epitope derived from the New York Esophageal Squamous Cell Carcinoma-1 (NY-ESO-1) cancer-testis antigen. It is utilized as an investigational peptide-based cancer vaccine designed to elicit a specific cytolytic T lymphocyte (CTL) response against tumor cells expressing the NY-ESO-1 antigen. NY-ESO-1 is a well-characterized cancer-testis antigen that is highly expressed in various malignancies, including cutaneous melanoma, but is absent in normal adult tissues except for germ cells in the testis. In clinical studies, such as a Phase 2 trial sponsored by Cliniques Universitaires Saint-Luc, NY-ESO-1b.A2 was administered to disease-free, HLA-A2-positive melanoma patients, often in combination with other melanoma-associated peptides and adjuvants like Montanide ISA 51VG and IMP321 (a LAG-3Ig fusion protein) to enhance the magnitude and kinetics of the immune response.
