NY-ESO-1b.A2 peptide vaccine is a peptide-based therapeutic cancer vaccine candidate developed by Cliniques Universitaires Saint-Luc. The vaccine contains the NY-ESO-1b peptide, an HLA-A2-restricted epitope (SLLMWITQC) derived from the New York Esophageal Squamous Cell Carcinoma-1 (NY-ESO-1) cancer-testis antigen. NY-ESO-1 is a highly immunogenic tumor-specific antigen expressed in various malignancies, including melanoma. The vaccine is designed to bind to the HLA-A2 molecule and present the NY-ESO-1 antigen to the immune system, thereby inducing a specific cytolytic T-lymphocyte (CTL/CD8+ T cell) response against NY-ESO-1-expressing tumor cells. In a Phase 2 clinical trial (NCT00365937) in Belgium, the vaccine was evaluated in disease-free HLA-A2-positive cutaneous melanoma patients as part of a multi-peptide vaccine regimen consisting of eight melanoma-associated HLA-A2-restricted peptides, administered with or without adjuvants such as Montanide ISA 51 and/or IMP321.
