NY-ESO TCR refers to a family of adoptive cell therapies in which autologous T lymphocytes are genetically engineered to express a T cell receptor (TCR) specific for the NY-ESO-1 (CTAG1B) cancer-testis antigen. NY-ESO-1 is a highly immunogenic antigen expressed on a range of tumors, including synovial sarcoma, melanoma, multiple myeloma, and triple-negative breast cancer, and not on normal tissues[6]. The engineered T cells recognize a defined peptide epitope from NY-ESO-1 (commonly NY-ESO-1 157–165) in the context of an MHC molecule (frequently HLA-A*02:01), bind tumor cells by this antigen, and mediate tumor cell lysis[3][6][7]. Some constructs (such as TBI-1301) use an affinity-enhanced receptor and include silencing of the endogenous TCR to improve surface expression and lower mispairing risk[3]. These therapies are generally administered by intravenous infusion following lymphodepleting chemotherapy[2][3].
