Overview

**NYESO SCT** is an investigational, autologous NY-ESO-1-directed adoptive cellular therapy developed by the Jonsson Comprehensive Cancer Center for NY-ESO-1-positive advanced solid tumors, including sarcoma. The treatment comprises separately administered peripheral blood stem cells engineered with a lentiviral vector encoding an NY-ESO-1-specific T-cell receptor and sr39TK safety gene, followed by peripheral blood mononuclear cells engineered with a retroviral vector encoding the NY-ESO-1-specific T-cell receptor. Following myeloablative busulfan and fludarabine conditioning, the engineered cell products are infused intravenously and supported with aldesleukin. The engineered T cells are intended to recognize NY-ESO-1 peptide presented by HLA-A*02 and mediate cytotoxic immune elimination of antigen-positive tumor cells.
