NYESO T cells is an autologous adoptive T-cell therapy designed to target the cancer-testis antigen NY-ESO-1 (New York Esophageal Squamous Cell Carcinoma-1) in the context of HLA-A2. Developed by The Christie NHS Foundation Trust, the therapy involves harvesting a patient's own T cells, genetically engineering them to express a T-cell receptor (TCR) specific to NY-ESO-1, and re-infusing them following lymphodepleting chemotherapy. It was evaluated in a Phase II clinical trial (EudraCT 2012-005327-33) as part of the EU-funded ATTACK project for patients with advanced oesophagogastric (gastroesophageal) cancer. The trial was prematurely terminated after only two patients were treated due to safety concerns (including a fatal case of bone marrow failure in one patient), and development of this specific therapy has been discontinued.
