NYVAC is a highly attenuated, replication-deficient vaccinia virus vector derived from the Copenhagen strain. It was engineered by the precise deletion of 18 open reading frames (ORFs) from the parental viral genome, which significantly reduces its virulence and host range while preserving its ability to induce potent immune responses. NYVAC serves as a versatile vaccine platform for expressing heterologous antigens from various infectious agents, such as HIV-1, Plasmodium falciparum, and Japanese encephalitis virus, as well as tumor-associated antigens. It is particularly effective in heterologous prime-boost regimens, often used as a boost following a DNA vaccine prime to elicit broad and durable T-cell and B-cell mediated immunity. The vector is classified as a poxvirus-based biologic vaccine vector that functions as an immunostimulant.
