Overview

NYVAC-HIV-PT4 is a recombinant, replication-deficient poxvirus (NYVAC) vector-based HIV vaccine candidate. It is engineered to express HIV-1 antigens and is used as part of multicomponent prime-boost regimens for the prevention of HIV infection. The vaccine aims to induce both cellular and humoral immune responses against HIV, particularly targeting the envelope (Env) glycoprotein gp120, which plays a key role in viral entry into host cells. In clinical trials, NYVAC-HIV-PT4 has been administered alone or in combination with DNA-based vaccines and/or purified Env protein boosts (such as AIDSVAX B/E), with the goal of enhancing antibody responses—especially those directed at the V1/V2 region of Env, which are associated with reduced risk of infection[1][7]. The vaccine has shown immunogenicity by eliciting cross-clade binding antibodies, neutralizing antibodies, and antibody-dependent cell-mediated cytotoxicity[1][7].
