NYVAC Nat-B env is an investigational HIV vaccine candidate that uses the highly attenuated vaccinia virus strain NYVAC as a viral vector to deliver the HIV-1 envelope (Env) gene from clade B (Nat-B). The vaccine is designed to induce immune responses, particularly targeting the HIV-1 Env protein, which is critical for viral entry into host cells. It has been studied in prime-boost regimens, often following DNA priming with a similar antigen and followed by boosting with either NYVAC vectors or co-administered Env protein. The goal of this approach is to elicit both humoral (antibody-mediated) and cellular immune responses against HIV-1, including polyfunctional CD4+ and CD8+ T cell responses as well as neutralizing antibodies. Clinical trials have shown that vaccines containing the NYVAC vector can induce strong immunogenicity but may be associated with more frequent reactogenicity compared to DNA-based vectors[2][3][4][5]. This candidate remains under clinical investigation for its safety and ability to generate protective immunity against HIV infection.
