Overview

NYX-2925 is a novel small molecule and new chemical entity developed by Aptinyx that acts as a positive allosteric modulator (PAM) of the N-methyl-D-aspartate receptor (NMDAR). It functions as a co-agonist to glutamate at the NMDAR and partially activates these receptors at low concentrations of endogenous agonists such as glycine or D-serine. This modulation influences neural pathways relevant to chronic pain processing. NYX-2925 has been investigated primarily for treatment of chronic pain conditions including painful diabetic peripheral neuropathy (DPN) and fibromyalgia. It crosses the blood-brain barrier and has demonstrated safety and tolerability in early clinical trials without dissociative side effects. Despite promising pharmacodynamic activity in humans consistent with its mechanism on NMDAR-mediated pathways, it failed to meet primary efficacy endpoints in Phase IIb trials for DPN and fibromyalgia but remains under investigation for potential therapeutic benefit in other neurological disorders[1][3][4][5][6][8].
