Overview

NYX-783 is a novel, oral small molecule that acts as a positive allosteric modulator of N-methyl-D-aspartate (NMDA) receptors, with selectivity for the GluN2B subunit. It was developed by Aptinyx for the treatment of central nervous system disorders, primarily post-traumatic stress disorder (PTSD) and opioid use disorder (OUD). Preclinical studies demonstrated that NYX-783 enhances NMDA receptor-mediated currents in both glutamatergic and GABAergic neurons in the medial prefrontal cortex, leading to improved fear extinction and reduced spontaneous recovery of fear memories—mechanisms relevant to PTSD. The drug showed promising safety and tolerability profiles in early clinical trials. The U.S. FDA granted Fast Track designation for its development in PTSD[1][4][6][7][8].
