oAdDR18 is an oncolytic adenovirus engineered to express a decoy-resistant variant of interleukin-18 (DR18). Interleukin-18 (IL-18) is a potent proinflammatory cytokine that stimulates interferon-gamma (IFN-γ) production and enhances the activity of T cells and natural killer (NK) cells. However, the therapeutic efficacy of recombinant IL-18 is limited by the endogenous decoy receptor, IL-18 binding protein (IL-18BP), which neutralizes its activity. DR18 is designed to be resistant to IL-18BP binding while maintaining its affinity for the IL-18 receptor. By delivering DR18 via an oncolytic adenovirus, oAdDR18 combines direct viral-mediated tumor lysis with localized, sustained expression of decoy-resistant IL-18, thereby overcoming immunosuppression in the tumor microenvironment and promoting robust local and systemic antitumor immune responses.
