oAdwtIL-18 is a preclinical-stage oncolytic adenovirus engineered to express wild-type interleukin-18 (wtIL-18). Developed by researchers at Nanjing University, this cytokine-armed oncolytic virus is designed to selectively replicate in and lyse cancer cells while locally delivering wtIL-18 to the tumor microenvironment. The virus features a modified adenovirus backbone with an Ad5/3 chimeric fiber to enhance tumor targeting and cell entry via desmoglein-2 (DSG2) and CD46, bypassing the need for the coxsackievirus-adenovirus receptor (CAR). Once expressed, wtIL-18 stimulates T helper 1 (Th1) and natural killer (NK) cells, promoting the production of interferon-gamma (IFN-γ) and enhancing antitumor immunity. In preclinical studies, oAdwtIL-18 has been evaluated in syngeneic mouse models of colorectal cancer, melanoma, and breast cancer, demonstrating significant tumor growth inhibition, although its efficacy is limited by the endogenous decoy receptor IL-18 binding protein (IL-18BP) compared to decoy-resistant variants like oAdDR18.
