OAP-103

OAP-103 is a topical gel formulation of dapansutrile (also known as OLT1177), a selective small molecule inhibitor of the NLRP3 (NOD-like receptor protein 3) inflammasome. Developed by Olatec Therapeutics, OAP-103 is designed to provide localized anti-inflammatory effects for the treatment of pain associated with osteoarthritis, particularly of the knee. By specifically inhibiting the NLRP3 inflammasome, the drug prevents the activation of caspase-1 and the subsequent processing and release of the pro-inflammatory cytokines interleukin-1β (IL-1β) and interleukin-18 (IL-18). These cytokines are central mediators in the inflammatory cascade that drives joint pain and cartilage degradation in osteoarthritis. The topical delivery of OAP-103 aims to achieve therapeutic concentrations at the site of inflammation while minimizing systemic exposure and potential off-target effects.
