OAV SG655-mGMP is an engineered **oncolytic adenovirus** designed to target and eliminate cancer stem cells, particularly in **hepatocellular carcinoma stem cells (Hep3B-C)** and **teratoma stem cells (ECCG5)**. It is armed with two transgenes: **11R-P53** (a tumor suppressor protein p53 fused with a cell-penetrating 11R peptide to enhance intracellular delivery and efficacy) and **murine granulocyte-macrophage colony-stimulating factor (mGM-CSF)** (an immune stimulator that recruits immune cells to the tumor microenvironment). The virus uses the human telomerase reverse transcriptase (hTERT) promoter for tumor-selective expression of E1A, mGM-CSF under the MCMV promoter, and 11R-P53 in the E3 region. The combination augments both direct tumor suppressor effects (via p53) and anti-tumor immunity (via GM-CSF), thus exerting **synergistic anti-cancer activities** through growth inhibition, apoptosis induction, and immune cell recruitment in preclinical models[1][3][5].
