OB-3 is a synthetic peptide fragment of the human obesity protein, leptin, specifically encompassing residues 116–122. Unlike the full-length leptin protein, OB-3 is a small, stable peptide that exhibits significantly improved permeability across the blood-brain barrier. It acts as a leptin receptor agonist, mimicking the weight-reducing and insulin-sensitizing effects of the native hormone. Developed primarily at Albany Medical College, OB-3 has demonstrated efficacy in preclinical models for the treatment of obesity and type 2 diabetes by reducing food intake, decreasing body weight, and improving glucose tolerance. The inclusion of a D-amino acid substitution (D-Leu-4) enhances its metabolic stability and biological activity compared to the native sequence.
