OB1

OB1 (also known as OB-001) is a stimuli-selective monoclonal antibody targeting the Transient Receptor Potential Vanilloid 1 (TRPV1) ion channel, developed by Oblique Therapeutics. TRPV1 is a well-validated target for pain management, but previous small-molecule antagonists failed in clinical development due to significant side effects, including systemic hyperthermia and impaired heat sensation. OB1 utilizes Oblique's proprietary Ab-Cross platform to achieve stimuli-selective inhibition, specifically blocking heat- and capsaicin-induced activation of the channel while sparing its response to protons. This selective mechanism is intended to provide potent analgesia for chronic and neuropathic pain without disrupting normal thermoregulation. In 2021, Oblique Therapeutics entered into a significant R&D collaboration and option agreement with an unnamed top-20 pharmaceutical company to advance the program as a potential non-opioid alternative for pain management.
