**Obatoclax + rituximab** is an investigational combination therapy studied primarily for hematological malignancies, such as chronic lymphocytic leukemia and B-cell lymphomas. **Obatoclax** is a small molecule BH3 mimetic that antagonizes anti-apoptotic BCL-2 family proteins (including Mcl-1, Bcl-XL, and Bcl-w), promoting cancer cell apoptosis. **Rituximab** is a monoclonal antibody targeting CD20 on B lymphocytes, inducing cell death via immune mechanisms such as antibody-dependent cellular cytotoxicity (ADCC), complement-mediated cytotoxicity (CMC), and direct apoptosis signaling. Obatoclax has shown the ability to augment rituximab-mediated ADCC and enhance anti-tumor effects in both rituximab-sensitive and -resistant models. Clinically, the combination has demonstrated promising activity in relapsed/refractory chronic lymphocytic leukemia and B-cell lymphoma, with acceptable tolerability profile in early-phase trials[1][2][4].
