Overview

AUTO1-SL2 (obeldescabtagene autoleucel) is an autologous CD19-targeted chimeric antigen receptor (CAR) T-cell therapy developed by Autolus Therapeutics. It is uniquely engineered with a "fast-off" rate CD19 binding domain, designed to mimic the natural kinetics of physiological T-cell receptor interactions. This kinetic profile is intended to allow for high levels of T-cell expansion and persistence while minimizing excessive cytokine release, potentially improving the safety profile compared to first-generation CAR-T therapies. While the product is approved for B-cell acute lymphoblastic leukemia under the brand name Aucatzyl, the AUTO1-SL2 designation is specifically used for its clinical development in B-cell mediated autoimmune diseases, including systemic lupus erythematosus (SLE) and lupus nephritis (LN). By depleting the CD19-positive B-cell population, the therapy aims to eliminate pathogenic B-cells and provide a durable immune reset for patients with refractory lupus.
