Obetinib (DTRM-031) is an orally bioavailable, small-molecule inhibitor of Bruton's tyrosine kinase (BTK). Developed by DTRM Biopharma, it is designed to treat various B-cell malignancies by blocking the B-cell receptor (BCR) signaling pathway, which is critical for the survival and proliferation of malignant B cells. Obetinib is particularly noted for its potential in treating central nervous system (CNS) lymphomas, often evaluated in combination with other therapies such as CAR-T cells (e.g., regorafenapyr), autologous hematopoietic stem cell transplantation (ASCT), and checkpoint inhibitors (e.g., sintilimab). It is currently being investigated in clinical trials for primary and relapsed/refractory CNS diffuse large B-cell lymphoma (DLBCL).
