OBI-201 is a preclinical-stage bispecific antibody-drug conjugate (ADC) developed by OBI Pharmaceuticals that simultaneously targets TROP2 and HER2. It is constructed using OBI's proprietary GlycOBI® platform, which utilizes the dual-function enzyme EndoSymeOBI® and the HYPrOBI® linker to conjugate a topoisomerase I inhibitor payload. By targeting two distinct tumor-associated antigens, OBI-201 is designed to overcome tumor heterogeneity and low antigen expression, potentially improving binding avidity and payload delivery while reducing off-target toxicity. In preclinical models of drug-resistant breast cancer with low HER2 expression, OBI-201 demonstrated superior anti-tumor activity compared to single-target ADCs.
