OBI-221 is a first-in-class bispecific dual-payload antibody-drug conjugate (BsDpADC) developed by OBI Pharmaceuticals. It simultaneously targets the cMET (Hepatocyte Growth Factor Receptor) and HER3 (Human Epidermal Growth Factor Receptor 3) receptors, which are often co-expressed and implicated in resistance to EGFR-targeted therapies. Utilizing the proprietary GlycOBI DUO® platform and HYPrOBI® linker technology, OBI-221 delivers two distinct, synergistic cytotoxic payloads directly to tumor cells. This dual-targeting and dual-payload approach is designed to overcome tumor heterogeneity and bypass common resistance mechanisms in solid tumors such as non-small cell lung cancer (NSCLC), colorectal cancer, and gastric cancer.
