OBI-3424 is a first-in-class, small-molecule chemotherapeutic prodrug designed to be selectively activated by the enzyme aldo-keto reductase family 1 member C3 (AKR1C3), which is overexpressed in certain tumor types. Upon intravenous administration, AKR1C3 converts OBI-3424 into a potent DNA alkylating agent that induces DNA cross-linking and subsequent cancer cell death. This targeted activation aims to minimize toxicity in normal tissues while maximizing cytotoxicity in tumors with high AKR1C3 expression. The drug has been investigated primarily for relapsed/refractory T-cell acute lymphoblastic leukemia (T-ALL), liver cancer, pancreatic cancer, and other solid tumors[1][5][7][9].
