Overview

OBI-833 is a carbohydrate-based therapeutic cancer vaccine designed as an active immunotherapy. It consists of the tumor-associated carbohydrate antigen Globo H conjugated to CRM197 (a non-toxic mutant of diphtheria toxin) and is administered with the adjuvant OBI-821 to enhance immune response. The mechanism involves stimulating the immune system to generate anti-Globo H antibodies (IgM and IgG), leading to complement-dependent cytotoxicity (CDC) and antibody-dependent cell-mediated cytotoxicity (ADCC) against tumor cells expressing Globo H. Developed by Academia Sinica and further advanced by OBI Pharma, it targets epithelial cancers that express the Globo H antigen, including non-small cell lung cancer (NSCLC) and biliary tract cancers such as cholangiocarcinoma[2][4][5][6][8][9].
