OBI-999 is a novel, first-in-class antibody-drug conjugate (ADC) designed to target cancer cells expressing the Globo H antigen, which is present in up to 15 types of epithelial cancers. The drug consists of a humanized monoclonal immunoglobulin G1 antibody (OBI-888) that specifically binds to the tumor-associated antigen Globo H and is conjugated via proprietary linker technology to monomethyl auristatin E (MMAE), an ultrapotent antimitotic agent. Upon binding to Globo H on cancer cells, the ADC is internalized and releases MMAE intracellularly, leading to inhibition of tubulin polymerization, cell cycle arrest, and apoptosis. OBI-999 has demonstrated dose-dependent tumor growth inhibition in preclinical models for breast, gastric, pancreatic, lung, colorectal, and esophageal cancers. It was developed by OBI Pharma and has received orphan drug designation for gastric and pancreatic cancer[1][2][3][4][5][6][8].
