**obinutuzumab + bendamustine + prednisone** is a multi-agent chemotherapy regimen used mainly for the treatment of **follicular lymphoma** and other B-cell lymphoid malignancies.\ - **Obinutuzumab** is a humanized, glycoengineered **type II anti-CD20 monoclonal antibody** that induces cell death primarily through antibody-dependent cellular cytotoxicity (ADCC), antibody-dependent cellular phagocytosis (ADCP), and direct cell death by non-apoptotic mechanisms[1][3][4][5][8].\ - **Bendamustine** is an **alkylating agent** (small molecule) causing DNA cross-linking, leading to apoptosis and cell death.\ - **Prednisone** is a **synthetic glucocorticoid corticosteroid** with immunosuppressive and anti-inflammatory activity, used to reduce infusion-related reactions and for anti-tumor effects[1][2][3][7][8].\ Obinutuzumab plus bendamustine, with or without prednisone, is administered intravenously and orally for induction treatment over several cycles. In clinical studies, prednisolone (or equivalent doses of prednisone) has been shown to enhance the ADCC sensitivity to obinutuzumab and to improve anti-tumor effects by mechanisms including upregulation of CD20 and apoptosis-inducing proteins[1][3]. The combination is effective against obinutuzumab-resistant clones of lymphoma cells.\ Main indications include previously untreated or relapsed/refractory follicular lymphoma and chronic lymphocytic leukemia.
