Overview

Obinutuzumab + lenalidomide is a combination therapy used for follicular lymphoma and other indolent B-cell non-Hodgkin lymphomas. Obinutuzumab is a type II humanized anti-CD20 monoclonal antibody that mediates B-cell killing by enhancing antibody-dependent cellular cytotoxicity, phagocytosis, and direct cell death. Lenalidomide is an oral immunomodulatory small molecule that exerts anti-tumor activity by direct effects on malignant cells, enhancement of T-cell and natural killer cell function, and modulation of cytokine production. Lenalidomide also increases antibody-dependent cell-mediated cytotoxicity, particularly when paired with anti-CD20 antibodies. The combination has demonstrated high efficacy, a favorable safety profile, and is considered a promising chemotherapy-free regimen for relapsed/refractory and untreated follicular lymphoma.
