Obinutuzumab + pixantrone is a combination therapy evaluated for the treatment of relapsed or refractory diffuse large B-cell lymphoma (DLBCL) and other aggressive lymphomas. Obinutuzumab is a type II glycoengineered anti-CD20 monoclonal antibody that targets the CD20 antigen on B cells, leading to cell death through direct cytotoxicity and immune-mediated mechanisms. Pixantrone is an aza-anthracenedione antineoplastic agent that acts primarily as a DNA intercalator and topoisomerase II inhibitor, causing double-stranded DNA breaks and cell death. The combination has shown clinical activity in heavily pretreated patients with relapsed/refractory DLBCL, but failed to meet its primary endpoint in phase II trials. Both agents have demonstrated individual benefit in aggressive lymphomas; their combination aims to improve outcomes for patients with limited options[3][4][7].
