This is a combination therapy of four anticancer drugs: - **Obrixtamig (HPN328, BI 764532):** A novel bispecific T-cell engager (TCE) antibody designed to target delta-like ligand 3 (DLL3), which is highly expressed in small cell lung cancer (SCLC) and other high-grade neuroendocrine cancers. Obrixtamig simultaneously binds DLL3 on tumor cells and CD3 on T cells, redirecting and activating T cells to kill DLL3-positive cancer cells. - **Cisplatin:** A platinum-based chemotherapy that forms DNA crosslinks, disrupting DNA synthesis and function, leading to apoptosis (cell death) in rapidly dividing cancer cells. - **Etoposide:** A topoisomerase II inhibitor that prevents DNA unwinding and replication, leading to DNA breaks and cell death. - **Durvalumab:** A human monoclonal antibody targeting programmed death ligand 1 (PD-L1), thereby blocking the PD-L1/PD-1 pathway and enhancing T cell immune response against tumor cells (immune checkpoint inhibitor). This combination is investigational for DLL3-positive small cell lung cancer and other neuroendocrine carcinomas, typically as part of advanced-line or clinical trial regimens[1][2][6].
