Drug intelligence / Profile preview
Obrixtamig + pumitamig is an investigational combination regimen being evaluated for the treatment of extensive-stage small cell lung cancer (ES-SCLC). The combination consists of obrixtamig (BI 764532), Boehringer Ingelheim's bispecific DLL3/CD3 T-cell engager, and pumitamig (BNT327/BMS-986545), a PD-L1/VEGF-A bispecific antibody jointly developed by BioNTech and Bristol Myers Squibb. Obrixtamig redirects CD3-positive T-cells to kill tumor cells expressing delta-like ligand 3 (DLL3), a protein highly expressed in SCLC and other neuroendocrine tumors. Pumitamig simultaneously blocks programmed death-ligand 1 (PD-L1) to restore T-cell activity and neutralizes vascular endothelial growth factor A (VEGF-A) to inhibit tumor angiogenesis and alleviate the immunosuppressive tumor microenvironment. Boehringer Ingelheim and BioNTech are evaluating the safety, tolerability, and efficacy of this dual-bispecific combination in a Phase Ib/II clinical trial (NCT07843940, Dareon-Rosetta35) for patients with previously treated or treatment-naive ES-SCLC.
