Overview

OC-1d is a preclinical dual-targeting CAR-T cell therapy developed by OneChain Immunotherapeutics for the treatment of T-cell acute lymphoblastic leukemia (T-ALL) and T-cell lymphoblastic lymphoma (T-LL). The therapy is engineered to simultaneously target two specific proteins, CD1a and CCR9, which are co-expressed on leukemic cells in a majority of T-ALL patients but are not significantly present on healthy T lymphocytes or hematopoietic stem cells. By targeting these specific antigens, OC-1d aims to induce potent anti-leukemic activity while avoiding the T-cell fratricide and subsequent T-cell aplasia often associated with pan-T-cell CAR-T therapies (e.g., those targeting CD5 or CD7). The dual-targeting approach is further designed to prevent immune escape by ensuring efficacy even if one of the target antigens is downregulated.
