Drug intelligence / Profile preview
OCA-B inhibitors are a class of therapeutic agents designed to target and inhibit the activity of the transcription co-regulator OCA-B (also known as POU2AF1 or OBF-1) and its interaction with the transcription factor Oct1 (POU2F1). OCA-B is highly expressed in B cells and is also expressed at lower levels in T cells, where it plays a critical role in CD4+ memory recall and autoimmune responses, but is dispensable for primary immune responses. Inhibition of OCA-B has shown therapeutic potential in autoimmune diseases, such as type 1 diabetes (T1D) and multiple sclerosis (MS), as well as in preventing checkpoint blockade-induced immune-related adverse events (irAEs) like PD-1 inhibitor-induced diabetes, without compromising anti-tumor immunity. Both peptide-based inhibitors (such as JumOCA) and small molecule inhibitors (such as DB1033 and DB2232) have been developed as proof-of-concept agents.
