Overview
Ocadusertib is an orally available, potent, and selective small molecule inhibitor of receptor-interacting serine/threonine-protein kinase 1 (RIPK1). It acts as an allosteric inhibitor, blocking RIPK1-mediated inflammatory cell death (necroptosis) and associated inflammatory responses. Preclinical studies demonstrated that ocadusertib inhibits RIPK1 enzymatic activity with high potency and prevents necroptosis-induced inflammation in various models. In clinical trials, it has shown a favorable safety profile and pharmacokinetics suitable for once-daily dosing. Ocadusertib is being developed primarily for the treatment of autoimmune and inflammatory diseases, with its lead indication being moderately to severely active rheumatoid arthritis[1][2][5][7].
