Overview

OCT1004 (uHAP 1) is a novel small molecule prodrug developed by OncoTherics for the treatment of solid tumors, including pancreatic ductal adenocarcinoma (PDAC) and brain cancers such as glioblastoma multiforme (GBM) and diffuse intrinsic pontine glioma (DIPG). It is designed as a dual-activated prodrug, triggered by both tumor hypoxia and ultrasound, which converts the non-cytotoxic precursor into an active topoisomerase II (TOPO 2) inhibitor specifically within the tumor microenvironment. This targeted activation mechanism aims to overcome resistance in hypoxic tumor regions while minimizing systemic toxicity. OCT1004 is engineered for deep tissue penetration and is capable of crossing the blood-brain barrier, making it a candidate for combination therapies with radiotherapy and chemotherapy (e.g., temozolomide).
