Drug intelligence / Profile preview
Octapeptin C4-1 is a synthetic, non-hydroxylated cyclic lipopeptide analog of octapeptin C4 being developed as a next-generation antibacterial agent. It is designed to target and disrupt bacterial membranes, circumventing resistance mechanisms associated with colistin and other polymyxins. The drug's mechanism of action involves initial electrostatic interactions between its polar/charged groups and the bacterial membrane, followed by the insertion of its hydrophobic tail into the lipid bilayer, leading to membrane disruption. Octapeptin C4-1 has demonstrated broad-spectrum bactericidal activity against both Gram-negative and Gram-positive pathogens, including colistin-resistant strains, with low cellular toxicity in preclinical models.
