Octreotide is a synthetic octapeptide analog of the natural hormone somatostatin. It acts as a more potent and longer-acting inhibitor of growth hormone, glucagon, and insulin than endogenous somatostatin. Octreotide binds with high affinity to somatostatin receptor subtype 2 and subtype 5, and with moderate affinity to subtype 3. Its primary mechanism involves inhibition of the secretion of several hormones (including growth hormone, insulin, glucagon, gastrin, vasoactive intestinal peptide), reduction in gastrointestinal motility and secretions, vasoconstriction in blood vessels (reducing portal pressure), and suppression of pituitary growth hormone release. Clinically it is used for the treatment of acromegaly (excess growth hormone production), symptomatic control in carcinoid tumors (to reduce diarrhea/flushing), vasoactive intestinal peptide-secreting tumors (VIPomas), and other neuroendocrine tumors[1][4][5][6][7].
