Octreotide is a synthetic octapeptide that mimics the natural hormone somatostatin, acting as a more potent inhibitor of growth hormone (GH), glucagon, and insulin. It is primarily used to treat acromegaly, severe diarrhea and flushing associated with metastatic carcinoid tumors, and vasoactive intestinal peptide-secreting tumors (VIPomas). Octreotide exerts its effects by binding to somatostatin receptors—mainly subtypes 2 and 5—leading to decreased secretion of several hormones and reduced splanchnic blood flow[1][5][6][8]. DG3173 (also known as somatoprim) is a novel synthetic analogue of somatostatin with selectivity for receptor subtypes 2, 4, and 5. It effectively suppresses GH secretion in human pituitary adenoma cells—including some that are non-responsive to octreotide—and has lower insulin-suppressive activity compared to other analogues. This profile suggests potential clinical advantages in treating acromegaly or related disorders where glucose tolerance is a concern[2]. The combination of octreotide + DG3173 represents two distinct but mechanistically similar drugs targeting the same pathway via different receptor subtype affinities.
