Octreotide-decorated cetuximab-loaded polymeric nanoparticles are an experimental oral drug delivery system developed by researcher Ahmed A. H. Abdellatif for the targeted treatment of colorectal cancer. The formulation utilizes pH-sensitive polymers (such as Eudragit S100 or calcium alginate) to encapsulate cetuximab, a chimeric IgG1 monoclonal antibody that inhibits the epidermal growth factor receptor (EGFR). The surface of these nanoparticles is decorated with octreotide, a somatostatin analogue, which serves as a targeting ligand for somatostatin receptors (SSTRs) frequently overexpressed in colorectal tumors. This dual-targeting approach is intended to facilitate site-specific release in the colon and enhance cellular uptake by cancerous tissues while minimizing systemic exposure and degradation of the biologic in the upper gastrointestinal tract. The candidate is currently being investigated in Phase 1 clinical trials.
