Octreotide LAR is a synthetic, eight-amino-acid analogue of the natural hormone somatostatin, designed for prolonged release. It functions by acting as a somatostatin analogue, which limits beta-cell insulin secretion by inhibiting the G0 protein associated with the widening of the voltage-gated calcium channel. Additionally, it inhibits the secretion of gastric acid, pancreatic enzymes, and bile, prolongs intestinal transit time, and decreases gallbladder contractility. Octreotide LAR also suppresses the release of various pituitary and gastroenteropancreatic hormones, including growth hormone (GH), thyroid stimulating hormone (TSH), glucagon, cholecystokinin (CCK), vasoactive intestinal peptide (VIP), gastrin, and ghrelin. It is developed by companies such as Novartis Pharmaceuticals and is used in conditions like advanced gastroenteropancreatic neuroendocrine tumors, acromegaly, polycystic ovary syndrome, and obesity with insulin hypersecretion.
