Overview

OCU200 is a novel recombinant fusion protein biologic composed of two human proteins, tumstatin and transferrin, joined by a peptide linker. Tumstatin acts as the active component with anti-inflammatory and anti–vascular endothelial growth factor (anti–VEGF) properties by binding to integrin receptors (notably αVβ3), which are implicated in the pathogenesis of retinal vascular diseases. Transferrin targets the drug to retinal and choroidal tissues via transferrin receptor-mediated uptake on endothelial cells, enhancing tissue penetration and bioavailability compared to tumstatin alone. The combined mechanism aims to inhibit angiogenesis, reduce inflammation, decrease neovascularization, and lower vascular permeability—key factors in diabetic macular edema (DME), diabetic retinopathy (DR), and wet age-related macular degeneration (wet AMD). Developed by Ocugen based on technology from University of Colorado Anschutz Medical Campus, OCU200 is currently in Phase 1 clinical trials for DME administered via intravitreal injection[1][2][3][5][7][9][10].
