OD-003 is a preclinical-stage protein therapeutic developed by Odyssey Therapeutics that acts as a selective agonist of Tumor Necrosis Factor Receptor 2 (TNFR2). While TNFR1 is widely expressed and often associated with pro-inflammatory signaling and apoptosis, TNFR2 expression is more restricted, primarily found on regulatory T cells (Tregs), certain subsets of effector T cells, and endothelial cells. Activation of TNFR2 promotes the expansion, stabilization, and suppressive function of Tregs, which are critical for maintaining immune tolerance. By agonizing this receptor, OD-003 is designed to restore immune homeostasis in patients with autoimmune and inflammatory diseases. The program is currently being evaluated for the treatment of atopic dermatitis, systemic lupus erythematosus (SLE), and alopecia areata.
