OD-07656 is an oral small molecule drug under development by Odyssey Therapeutics for the treatment of inflammatory diseases, including ulcerative colitis, Crohn's disease, spondyloarthritis, and Blau syndrome. It acts as a receptor-interacting serine/threonine-protein kinase 2 (RIPK2) scaffolding inhibitor. By blocking RIPK2 scaffolding with XIAP, OD-07656 aims to inhibit downstream signaling pathways that activate proinflammatory cells in both the innate and adaptive immune systems. This mechanism reduces the production of key cytokines such as TNF, TL1A (TNF-like cytokine 1A), and IL-23—cytokines implicated in the pathology of inflammatory bowel disease (IBD). Preclinical data suggest OD-07656 has anti-inflammatory and anti-fibrotic effects and may be effective both as monotherapy or in combination with existing IBD treatments to overcome resistance[1][7][8].
