ODC-siRNA is a small interfering RNA (siRNA) designed to silence the expression of ornithine decarboxylase (ODC), the rate-limiting enzyme in the polyamine biosynthetic pathway. Polyamines are essential for cell growth and proliferation, and ODC is frequently overexpressed in various cancers, including melanoma. By targeting ODC mRNA for degradation via the RNA interference (RNAi) pathway, ODC-siRNA inhibits polyamine synthesis, leading to growth arrest and apoptosis in tumor cells. In preclinical studies, ODC-siRNA has demonstrated significant growth inhibitory effects on human melanoma cell lines, particularly when used in combination with other inhibitors like mTOR inhibitors (e.g., rapamycin).
