Overview

The ODC1 neoantigen peptide-pulsed dendritic cell vaccine is an autologous cellular immunotherapy developed at Duke University for the treatment of glioblastoma. The vaccine is prepared by loading (pulsing) a patient's own dendritic cells with a neoantigen peptide derived from Ornithine Decarboxylase 1 (ODC1), a protein involved in polyamine biosynthesis that is frequently dysregulated in brain tumors. Once administered, typically via intradermal injection, these dendritic cells present the ODC1 antigen to the immune system, aiming to stimulate a targeted CD8+ T-cell response against tumor cells. Research has also explored the use of systemic CCL3 as an adjuvant to enhance the migration of the vaccinated dendritic cells to lymph nodes, thereby improving the overall anti-tumor efficacy.
