ODI-2001 is a personalized therapeutic cancer vaccine regimen being developed by Centre Georges Francois Leclerc. It consists of a combination of a DNA neoantigen vaccine, a Modified Vaccinia virus Ankara (MVA) viral vector adjuvant, and the checkpoint inhibitor ipilimumab (an anti-CTLA-4 monoclonal antibody). The vaccine is designed to stimulate a patient-specific immune response against tumor-specific neoantigens. The DNA vaccine provides the genetic template for the neoantigens, while the MVA vector acts as a potent viral adjuvant to enhance the immune response. Ipilimumab works by blocking the CTLA-4 checkpoint, thereby sustaining T-cell activation and overcoming tumor-induced immunosuppression. It is currently being evaluated in Phase 1 trials for metastatic or locally advanced colorectal and pancreatic cancers.
