Overview

ODM-209 is an orally bioavailable, potent and selective small molecule inhibitor of cytochrome P450 side-chain cleavage enzyme (CYP11A1), also known as cholesterol side-chain cleavage enzyme. By inhibiting CYP11A1, it blocks the conversion of cholesterol to pregnenolone—the first and rate-limiting step in steroid hormone biosynthesis. This leads to suppression of steroid hormone production (including androgens and estrogens), which is relevant for the treatment of hormone-dependent cancers such as metastatic castration-resistant prostate cancer (mCRPC) and advanced breast cancer. Preclinical studies show that ODM-209 inhibits proliferation of hormone-positive tumor cells. The drug was developed by Orion and has been evaluated in phase 1/2 clinical trials for mCRPC and estrogen receptor-positive/HER2-negative advanced breast cancer[1][2][3][5][6].
