**ODN 1585** is a class A CpG oligodeoxynucleotide (ODN) that functions as a synthetic immunostimulatory agent designed for the activation of **Toll-like receptor 9 (TLR9)**, primarily in mice. It contains unmethylated CpG motifs, which mimic microbial DNA and elicit strong immune responses via TLR9 signaling. ODN 1585 is optimized to stimulate natural killer (NK) cell function, enhance CD8+ T-cell responses, induce dendritic cell migration, and promote a Th1-skewed immune response. It has demonstrated antitumor effects in several preclinical mouse models, particularly for melanoma and MHC class I-deficient tumors, where it mediates tumor regression by activating NK cells and cytotoxic T lymphocytes. Additionally, ODN 1585 can downregulate cyclic GMP-AMP synthase (cGAS) post-translationally in fibroblastic reticular cells (FRCs), involving recruitment of ubiquitin ligases such as ZNF598, thus suppressing the cGAS–STING signaling axis. Its primary application is as a research reagent or as an adjuvant in experimental immunotherapy, not as an approved pharmaceutical product[1][2][3][4][7][9].
