A combination therapy consisting of odronextamab, a bispecific monoclonal antibody targeting CD20 and CD3, and lenalidomide, an immunomodulatory small molecule. Odronextamab engages both T cells (via CD3) and malignant B cells (via CD20) to induce T cell-mediated cytotoxicity against B-cell non-Hodgkin lymphoma. Lenalidomide modulates the immune response and exhibits antineoplastic activity through cereblon-dependent mechanisms, enhancing cytotoxic immune cell activity and inhibiting tumor-promoting microenvironment interactions. The combination is under clinical investigation, primarily for relapsed/refractory follicular lymphoma (FL) and marginal zone lymphoma (MZL), subtypes of non-Hodgkin lymphoma, and the regimen is considered first-in-human as a combination.
